Abstract

Celem tej pracy była synteza i analiza nowych molekularnych makrocykli i klatek opartych na wiązaniu disulfidowym. Synteza funkcjonalnych architektur molekularnych stanowi intrygujące wyzwanie badawcze ze względu na liczne zastosowania takich struktur. Pierwsza część pracy została poświęcona syntezie i analizie pięciu dimerycznych klatek molekularnych rozpuszczalnych w wodzie. Przedstawiono pełny opis syntezy i analizę spektroskopową każdej klatki. Dodatkowo wykazano potencjał aplikacyjny otrzymanych struktur jako fluorescencyjne sensory wybranych lantanowców w wodzie. W drugiej części opisano metodę modyfikowania rozpuszczalności złożonych multidynamicznych klatek molekularnych. Modyfikacja bloków budulcowych o dodatkowy łańcuch boczny wprowadziła nową funkcjonalność do struktury całej klatki. W efekcie otrzymano i zbadano dwie nowe wieloskładnikowe klatki molekularne o zróżnicowanej polarności i właściwościach fluorescencyjnych. W ostatniej części pokazano oryginalne podejście do stymulowania biblioteki dynamicznej. W syntezie mikrofalowej otrzymano ditiolowy blok budulcowego, który zawiera grupy imidowe. Hydroliza in situ tych grup w warunkach zasadowych uwolniła nieoczekiwaną labilność konformacyjną komponentu. Zaowocowało to różnorodną strukturalnie biblioteką disulfidowych makrocykli. Opracowano metodę kontroli tego zjawiska poprzez modulację pH i przez dodatek DMSO.
The aim of this thesis was the synthesis and analysis of the new molecular macrocycles and cage-like systems based on reversible disulfide bonds. The synthesis of functional architectures continues to pose an intriguing challenge due to the numerous applications of such structures. The first part of this work deals with the synthesis and analysis of five different water-soluble dimeric molecular cages. A full description of the synthesis and analysis of each cage is provided. Additionally, the application potential of the obtained structures is presented as fluorescent sensors of selected lanthanides in water. The second part presents the solubility tuning in multi-dynamic molecular cages. By the structural modification of building blocks, the new functionality was introduced to the structure of the entire cage. That way, two new multi-component molecular cages with various polarity and fluorescent properties were obtained. The last part presents an unorthodox approach to stimulating a dynamic library. The use of microwave-assisted synthesis has resulted in a dithiol building block containing imide groups. The in situ hydrolysis of those imides under basic conditions caused unexpected conformational flexibility of this component that resulted in the library of disulfide macrocycles. The method of controlling this phenomenon by pH tuning and the addition of DMSO has been developed.
